Spin labels with different lengths of hydrophobic tails were tested to determine the influence of the length of the hydrophobic tail on the properties of liposomes. The compounds tested were DPH, DOPC, and a series of spin label derivatives of phosphatidylcholine with either methyl or ethyl chain. The liposomes were made in the presence of the spin label and in their absence. In the absence of the spin label, the liposomes were found to be stable and, above a critical concentration, the order parameters of the molecules in the lipids were measured by the electron paramagnetic resonance of the spin label. At lower concentrations, the molecules were found to move about freely in the liposomes. The movement of molecules in liposomes was found to be influenced by the length of the hydrophobic tail of the spin label. The length of the tail of the spin label was found to have a dual effect.
