We have previously isolated a factor, IL-2-dependent factor (IL-2DF), which induced a differentiation program in the human lymphoblastoid cell line IM-9. IL-2DF activity was highly induced during the first 3 h of treatment of the cells with the differentiation reagent TPA. We have cloned IL-2DF and have identified it as p65, a member of the nuclear factor-kappa B (NF-kappaB) family of transcription factors. We demonstrate here that p65, but not a nonhomologous human NF-kappaB1 (p50), was highly induced by TPA. TPA treatment of IM-9 cells led to increased transcriptional activity and histone H4 acetylation of a DNA-binding domain (C-terminal part) of human p65 in a TPA-concentration-dependent manner.
