The common triazole antifungal drugs fluconazole and itraconazole inhibit the protein histidine kinase Chk1p, a yeast protein involved in DNA synthesis and cell cycle control. Crystals of the complex of Chk1p with an inhibitory synthetic triazole (derived from fluconazole) were obtained and their structure was determined to 2.1 angstroms resolution using the multiple isomorphous replacement method with anomalous scattering. Two-dimensional classification of the structure revealed a number of well-defined binding sites within the asymmetric unit with very similar space groups, indicating a complex binding pattern at the specific Chk1p binding site of the triazole compound. The inhibitor binds preferentially to a site located in a large, square-shaped cavity, which encloses much of the N-terminal lobe of the protein. The triazole binds to a substantial number of residues that correspond to the ATP-binding site of the catalytic domain of other proteins. In addition to this specific interaction with the ATP-binding region of Chk1p, there are other interactions between the ATP-binding site and hydrophobic residues in the triazole structure. Structural evidence for the failure of the common triazole antifungals to inhibit the
