We propose to study the mechanism of ligand recognition by the heme-binding protein Myoglobin (Mb). Myoglobin is the prototypic member of the three-helix bundle family of heme-binding proteins. In its native state Mb is present in virtually every vertebrate muscle tissue where it plays an essential role in energy production. When ligand-free Mb binds heme it will dissociate from its binding site and exhibit a wide spectrum of ligand-induced conformational states. Mb is a challenging system because of the remarkable variety of conformations accessible to the protein when bound to various ligands. The complex conformational properties of Mb and its ability to control ligand-binding stoichiometry make Mb an ideal system to develop and test hypotheses about the details of ligand recognition by three-helix bundle proteins in general.
